Are you asking from a builder's standpoint? The level builder numbers the rooms automatically.

Is there any particular significance to the number of a room as to what position a given room has to all the rest of the rooms? I hope that makes some sort of sense.

Yes. When you start building a mansion, you pick a square to begin with. Each room is one square in the level builder. The level builder has a number for all the squares, so it keeps track of which squares you use and that's how each room gets its number.
